The project is directed towards a thorough study on the synthesis and physico-chemical investigation of modified oligonucleotides with isopolar, nonisosteric linkage - potential "antisense" drugs. By means of a complex set of measuring and computational methods, including NMR, UV, UV-CD and Raman spectroscopy, quantum-chemical calculations and computer simulations, the structural properties of the novel oligonucleotides and their compatibility with complementary chains of natural nucleic acids are investigated. Other characteristics needed for the biological effect, like the ability to penetrate to living cell or stability towards enzymes, are also determined. The aim is to find correlations among the chemical composition of the internucleotide linkage, the physico-chemical properties of the modified oligonucleotide, and its biological "antisense" efficiency. |
